Perbandingan Antara Kualitas DNA Daun Menggunakan Metode KIT (Promega) dan Metode Manual

Plant oil palm (Elaeis guineensis Jacq.) is one of the crops that occupy a position important in the sector of the plantation. Because the thing that is required presence information complete including information of molecular. Extraction of DNA is the stage of the first study of molecular were very influential to the quality of the isolation of DNA. Isolation of DNA among others must produce DNA without the presence of contaminants such as proteins and RNA. Because the use of manual methods in DNA isolation this time has never been done, so it is necessary the comparison between the methods for comparing the methods of the best in produce quality isolation of DNA. This research was conducted to determine the comparison of the quality of oil palm leaf DNA (Elaeis guineensis Jacq.), As well as the effectiveness of the KIT method (promega) and manual methods. This research was conducted at the Molecular Breeding Laboratory of PT. Bina Sawit Makmur (Sampoerna Agro) Palembang, on the date 1-31 The August 2019. Isolation of DNA leaves of the coconut palm is done on the type of leaf of coconut oil are still young by using the methods of manual and KIT (promega). The parameters observed in the form of quality DNA to see the percentage and purity of DNA. Result of the study showed that the dilution of DNA work leaves using methods KIT (promega) has the result of more good in quality, purity DNA produced in the method KIT (promega) ranges from 1.579 to 3.226 in the long-wave λ260/280  and 0.694 to 2.261 on a long-wave  λ260 / 230. Methods of manual, case is based on the results of purity that is generated on the isolation of DNA and efficiency of time that is needed in the construction, will be but costs are incurred far more expensive than the method manual.
