Pengujian Bakteri Golongan Coliform Menggunakan Metode Petrifilm Pada Sampel Air Sungai di Provinsi Sumatera Selatan

Coliform is a member of Gram-negative bacteria, rod-shaped, does not produce spores and can ferment lactose by forming gas bubbles in fermenting acid at a temperature of 37 ° C for a period of 48 hours. To be more efficient in determining Coliform, a Petrifilm is used which can show shades of red and blue. Approximately 97% of Escherichia coli produce betaglucuronidase which produces a blue precipitate. The purpose of this study was to determine the level of Coliform pollution in rivers in South Sumatra. This research is an experimental research, the design used is completely randomized design. It is known that from 5 samples of river water that have been tested there are 4 positive river samples containing total Coliform and only 1 sample is negative, while of the 5 samples there is no Escherichia coli bacteria found, for total coliform bacteria the highest is 700 CFU / mL in time. 48 hours incubation on the Pal river. From a physical point of view, river water contaminated with Coliform is cloudy and has a strong odor. The MR River (600 CFU / mL), the Banyu River (300 CFU / mL), the Lahat River (500 CFU / mL), and the Pal River (700 CFU / mL) contain total coliform bacteria that exceed the quality standard threshold of Permenkes No. 32 years 2017. So it is not suitable for use in daily activities, because this Coliform bacteria has a dangerous impact
